The Parsortix

The Angle PLC Parsortix is a small benchtop system using a patented micro-fluidic technology in the form of a disposable cassette to capture and then harvest circulating tumour cells (CTCs) from blood. The cassette captures CTCs and other types of cells based on their less deformable nature and larger size compared to other components of blood or other bodily fluids. The system is also often used to pre-enrich cells for the DEP-Array system. Parsortix-enriched cells can be cultured, used for FISH staining or downstream molecular/genomics applications.

The BRC flow core offers a full service on the Parsortix bookable via ilabs. Please book a full 4 h hour morning session (9 AM- 1 PM) or afternoon session (1 PM – 5 PM) for each cell sample to be run.

Users can chose from the following service options:

  1. Parsortix only, user provides stained or unstained blood sample
  2. DEPArray plus Parsortix and staining: user provides blood sample and antibodies

Users will bring their sample to the core and will obtain isolated cells for further manipulation. The core will provide disposable cassettes (included in price), consumables, instrumentation and manpower.

The DEPArray

The Silicon Biosystems DEPArray™ cell sorting platform uses a proprietary DEPArray™ cartridge to control the manipulation and collection of very rare cells. Researchers can analyze samples containing from a few to tens of thousands of cells. The single-use, microfluidic cartridge contains an array of individually controllable electrodes, each with embedded sensors. This circuitry enables the creation of dielectrophoretic (DEP) cages around cells. After imaging, individual cells of interest are gently moved to specific locations on the cartridge, e.g. for cell-cell interaction studies, or into the holding chamber for isolation and recovery.

The BRC DEPArray system allows image based selection of cells using 4 fluorescent channels (DAPI/FITC/PE/APC) and various morphological parameters with CellBrowserTM software. The system optics provide 10x magnification (0.64 micron/pixel resolution) and 20x magnification (0.32 micron/pixel resolution). Individual single cells as well as pools of cells can be selectively isolated for further cell culture or subsequent molecular/genomics applications.

The DEPArray is compatible with a wide variety of live or fixed rare cell suspensions so you can selectively sort and gain maximum information from enriched CTCs and samples with small cell loads and small tissue biopsies (e.g. fine needle aspirates, skin punches) to study the biological significance of rare cells in disease development, progression, and therapeutic response.
